Standard Operating Procedure for Operation and Calibration of UV Cabinet

1.0 Purpose:

To lay down a procedure for Operation and Calibration of UV Cabinet.

2.0 Scope:

The scope of this procedure is applicable for Operation and Calibration of UV Cabinet.

3.0 Responsibilities:

QC Personnel: To operate, clean and calibrate UV cabinet. And, to prepare the calibration report and certificate of calibration.

Section Head – QC / Designee: To check the calibration report and certificate of calibration.

Head QC/Designee: To approve the calibration report and certificate of calibation.

4.0 Definitions:

Not applicable

5.0 Procedure:

5.1 Ensure that the UV cabinet is clean before and after every operation.

5.2 Close the door properly before starting UV light.

5.3 Turn “OFF” UV light before placing or removing sample from UV chamber.

5.4 Check UV lamp is in working condition.

5.5 Ensure that the UV cabinet is within valid period of calibration.

5.6 Maintenance / Repairs:

5.6.1 When the instrument do not comply with the requirement / acceptance criteria specified, the instrument should be labelled “OUT OF CALIBRATION”. Instrument shall be repaired / serviced and recalibrated as per Calibration SOP.

5.7 Do not attempt to look directly at the lighted source.

5.8 Use Personal Protective Equipment (PPE) like Safety Goggles, Hand Gloves, while performing the activity on UV cabinet.

5.9 Handling of the sample under UV light should be done using opaque hand gloves to protect the hands from UV radiation.  Cotton, Leather or rubber gloves are suitable.

5.10 Operating Procedure :

5.10.1 Turn “ON” the mains of the UV cabinet.

5.10.2 Switch “ON” the visible light if required.

5.10.3 Open the door of the UV cabinet by pulling forward its knob with enough force to overcome its magnetic latch. Introduce and position the sample to be   tested into the UV cabinet and close the door.  Switch “OFF” the VISIBLE LIGHT and turn “ON” the switch of the required UV light-UV-short 254 nm or UV-long 365 nm needed for the particular inspection.

5.10.4 Examine the Sample.

5.10.5 After examine the sample, turn “OFF” the UV light and turn “ON” the visible light. Open the door.

5.10.6 Remove the sample.

5.10.7 Insert the next sample if required.

5.10.8 Turn “OFF” the visible light.

5.10.9 Turn “OFF” the mains.

5.11 Cleaning :

5.11.1 Clean the internal of UV cabinet with wet cloth. Followed by dry cloth.

5.11.2 Wipe out outer surface with help of dry clean cloth.

5.11.3 Ensure the proper cleaning of UV cabinet.

5.12 Calibration :

5.12.1 Weigh 0.2 g of sodium salicylate and transfer in 100 ml volumetric flask. Dissolve and dilute upto the mark with 95 % ethanol.

5.12.2 Weigh 0.04 g of sodium salicylate and transfer in 100 ml volumetric flask. Dissolve and dilute upto the mark with 95 % ethanol.

5.12.3 Take a chromatographic plate coated with silica gel G.

5.12.4 Apply two times 5 µL spot of 0.04 % w/v sodium salicylate solution having diameter about 5 mm. [a]

5.12.5 Similarly apply two times 5 µL spot of 0.2 % w/v sodium salicylate solution having diameter about 5 mm. [b]

5.12.6 Allow drying in air for 5 minutes.

5.12.7 Switch “ON” the mains.

5.12.8 Open the door of the cabinet.

5.12.9 Place the chromatographic plate in UV cabinet.

5.12.10 Select the wavelength 254 nm by pressing “UV-SHORT 254 nm” knob.

5.12.11 Examine Spot [a] in a position normal to the radiation of UV lamp through view glass.

5.12.12 Select the wavelength 365 nm by pressing “UV-LONG 365 nm” knob.

5.12.13 Examine Spot [b] in a position normal to the radiation of UV lamp through view glass.

5.12.14 Prepare the calibration check report and check for meeting the acceptance criteria.

5.12.15 Update the status of UV cabinet as “CALIBRATED”.

5.12.16 If calibration is not satisfactory inform Manager QC and take corrective action.

6.0 Acceptance Criteria:

Calibration: The Lamp should be capable of revealing without doubt spots of (a) and (b) of sodium salicylate at 254 nm and 365 nm, respectively.

7.0 Frequency:

Operation: As and when required.

Cleaning: Before and after every operation.

Calibration: Quarterly – 5 days.

8.0 Format for recording:

8.1 Format for “Instrument Calibration Protocol”.
