Standard Operating Procedure for Operation and Cleaning of Vacuum and Pressure Pump
1.0 Purpose:
To lay down a procedure for Operation and Cleaning of Vacuum / Pressure Pump.
2.0 Scope:
scope of this procedure is applicable for Operation and Cleaning of Vacuum / Pressure Pump.
3.0 Responsibility:
QC Personnel: To Operate and Clean Vacuum/ Pressure Pump.
Head QC/Designee: To Monitor the Activity.
4.0 Definitions:
NA
5.0 Procedure:
5.1 Confirm the voltage of the electric supply.
5.2 Use 0.45µm filter paper or as specified in the respective specification for filtering the mobile phase or any solvent.
5.3 Release the vacuum before stopping the pump.
5.4 Do not run the vacuum pump without solvent.
5.5 Placement of Filter Paper:
• The filter assembly consists of a stainless steel funnel (filter holder) and a filtering flask.
• Pull the handle of the stainless steel funnel upwards and remove the lower part of the funnel.
• Place the filter paper on the lower part of the funnel and attach it back to the upper part by pulling the handle downwards.
5.6 Filtration:
• Switch “ON” the mains.
• Place the funnel on the filtering flask.
• Connect the filtering flask with the inlet tube, which is connected to the vacuum pump.
• Pour the mobile phase or any solvent into the funnel.
• Start the vacuum pump by pressing the power button on it.
• The mobile phase or solvent will get filtered through pressure and will be collected in the filtering flask. • The pressure gauge on the vacuum pump will show the pressure.
• After filtration is complete, disconnect the inlet tube from the filtering flask to release the vacuum. • Switch “OFF” the vacuum pump and remove the funnel.
• Collect the mobile phase or any solvent in an appropriate container.
• Switch “OFF” the mains.
5.7 Cleaning:
• Clean the vacuum pump with a dry, clean, lint-free cloth.
• Clean the filter assembly and filtering flask with purified water.
